چکیده

Abstract: Cryo-electron tomography (cryo-ET) enables visualization of protein complexes within their native cellular environment at molecular resolution. Most cells and all tissues, however, are too thick to be imaged directly by transmission electron microscopy (TEM). Overcoming this limitation requires the production thin biological sections called lamellae. The procedure obtain lamellae cells, either seeded or grown on grids, cryo-focused ion beam (cryo-FIB) milling samples. This method faces an additional challenge when dealing with tissues multicellular organisms, as these samples must high-pressure frozen, which embeds sample in a layer ice. Nonetheless, can still prepared from such extracting small volume transferring it receiver grid for lamella preparation, process lift-out. Here, we describe available workflows produce lift-out cryogenic conditions recent developments gas injection system (GIS)-free approaches transfer. These advances expand applications cryo-ET, enabling investigation whole organisms situ

A novel cryo-FIB lift-out procedure for cryo-TEM sample preparation

The focused-ion-beam (FIB) is the method of choice for site-specific sample preparation for Transmission Electron Microscopy (TEM) in material sciences. A lamella can be physically lifted out from a specific region of a bulk specimen with submicrometer precision and thinned to electron transparency for high-resolution imaging in the TEM. The possibility to use this tool in life sciences applica...

Cryo-electron tomography of rubella virus.

Rubella virus is the only member of the Rubivirus genus within the Togaviridae family and is the causative agent of the childhood disease known as rubella or German measles. Here, we report the use of cryo-electron tomography to examine the three-dimensional structure of rubella virions and compare their structure to that of Ross River virus, a togavirus belonging the genus Alphavirus. The ecto...

Cryo-electron Tomography of Whole Cells

Cryo-electron tomography was used for studying the molecular organization of the cytoplasm in intact cells. Flat areas of cells grown on EM grids and preserved by rapid freezing within milliseconds were imaged in 3D. In the resulting maps the selected morphological and structural features were analyzed at unprecedented detail, and frequently correlated to lightmicroscopy images. We portray here...

Cryo-electron tomography of vaccinia virus.

The combination of cryo-microscopy and electron tomographic reconstruction has allowed us to determine the structure of one of the more complex viruses, intracellular mature vaccinia virus, at a resolution of 4-6 nm. The tomographic reconstruction allows us to dissect the different structural components of the viral particle, avoiding projection artifacts derived from previous microscopic obser...

Dynamics of antibodies from cryo-electron tomography.

The issue of protein dynamics and its implications in the biological function of proteins are arousing greater and greater interest in molecular biology. In cryo-electron tomography experiments one takes several snapshots of a given biological macromolecule. In principle, a large enough collection of snapshots may then be used to calculate its equilibrium configuration in terms of the experimen...
